摘要:随机购买重庆地区市场上具有代表性的人参饮料,利用超高效液相色谱(Ultra Performance Liquid Chromatography,UPLC)分析其中的人参三醇皂苷Re、Rg1、Rg2、Rf和人参二醇皂苷Rb1、Rb2、Rc、Rd、Rb3的含量。仪器条件为:色谱柱:岛津Shim-pack GISS柱(100mm×2.1mm,1.9μm);流动相:A为0.1%甲酸溶液,B为乙腈,梯度洗脱;流速:0.4mL/min,进样量:20μL;检测波长:203nm。人参饮料中被检测的9种人参皂苷成分线性关系良好,相关系数(R2)值均在0.995以上;日内精密度在1.7%~2.2%之间,日间精密度在2.5%~3.3%之间,重复性与稳定性良好;9种人参皂苷待测物的加标回收率在93.4%~112.7%之间,相对标准偏差(Relative Standard Deviation,RSD)在0.4%~4.3%(n=6)之间。该方法测定人参皂苷含量方便快速、准确灵敏、重复性好,适用于饮料中多种皂苷类化合物含量的测定,为皂苷类化合物分析提供技术支撑。

Analysis of 9 Ginsenosides in Ginseng Beverages Based on UPLC Technology

Abstract: Representative ginseng beverages in the Chongqing market were randomly purchased, The contents of protopanaxatriol Re, Rg1, Rg2, Rf and protopanaxadiol Rb1, Rb2, Rc, Rd, Rb3 were analyzed by Ultra Performance Liquid Chromatography (UPLC). Instrument conditions: chromatographic column: Shimadzu Shim pack GISS column (100 mm×2.1 mm, 1.9 μm); Mobile phase: A is a 0.1% formic acid solution, B is acetonitrile, gradient elution; Flow rate:0.4 mL/min, injection volume:20 μL; Detection wavelength:203 nm. The linear relationship among the 9 ginsenoside components detected in ginseng beverages was good, with correlation coefficient R2 values all above 0.995. The within-day precision ranges from 1.7% to 2.2%, and the between-day precision ranges from 2.5% to 3.3%, indicating good repeatability and stability; The spiked recovery rates of 9 saponin analytes were between 93.4% and 112.7%, and the relative standard deviation (RSD) was between 0.4% and 4.3%(n=6). This method for determining the content of ginsenosides is convenient, fast, accurate, sensitive, and has good repeatability. It is suitable for the determination of various saponin compounds in beverages and provides technical support for the analysis of saponin compounds.
